Inner spokes run from the hub out to the rim on the side closer to the centerline of the wheel, and the right-hand/left-hand and inner/outer distinctions matter because a front wheel is laced with several different spoke lengths and angles depending on position. Getting the wrong one in the wrong hole will throw the lacing pattern off and can leave you short or with a spoke that won't seat right at the nipple.
